Sleep-promoting activity of medicinal plants containing quercetin-3-glucuronide in a pentobarbital-induced sleep model

Abstract
The purpose of this study was to evaluate the sleep-promoting activities of plant extracts by screening domestic plants with a high quercetin-3-glucuronide (Q3G) content. The Q3G (20 mg/kg) group had a significantly improved sleep latency time (p<0.001) and sleep duration time (p<0.05) compared to that of the normal group. In addition, Q3G has a high binding affinity to the benzodiazepine site of the GABAA receptor and thus promotes sleep through GABAergic action. Extracts of Filipendula multijuga Max. (14.10 mg/g), Cacalia firma Kom. (10.95 mg/g), and Persicaria perfoliate (45.37 mg/g) had higher Q3G contents compared to that of other domestic plants. The Persicaria perfoliate extract exhibited the highest sleep-promoting activity among the three extracts. Furthermore, it had a significantly higher polyphenol (p<0.05) and flavonoid (p<0.05) contents among the three extracts and showed excellent radical scavenging abilities. Therefore, the Persicaria perfoliate extract has great potential as a functional material for improving sleep disorders.
